Tissue-specific determinants of the adaptive NK cell response

چکیده

Abstract Natural killer (NK) cells are circulating innate lymphocytes that poised to respond rapidly upon encounter with cancer or virally-infected as well proinflammatory cytokines. In addition their early role in immune responses, it is now appreciated NK can also take on features of adaptive such antigen-specific clonal expansion and long-lived memory these processes best characterized following cytomegalovirus (CMV) infection mouse human. cell receptor recognition virally-encoded proteins drives CMV-specific cells, but the signals collectively give rise still not fully understood. Mouse (MCMV) systemic while exhibit cytokine-induced activation across multiple tissue sites infection, we were surprised find MCMV-specific occurs predominantly spleen. Transcriptional analysis indicated canonical non-canonical NF-kb components may be engaged a greater extent primed spleen, further identified TNFa signaling critical regulator both responses through engagement distinct downstream arms. Notably, TNFR2-deficient exhibited hyper-activation ultimately failed undergo strong proliferative burst observed These findings highlight central spleen facilitating innate-to-adaptive transition provide insight into how better generate immunity diverse settings. Tissue-Specific Education of Decidual NK Cells

During human pregnancy, fetal trophoblast cells invade the decidua and remodel maternal spiral arteries to establish adequate nutrition during gestation. Tissue NK cells in the decidua (dNK) express inhibitory NK receptors (iNKR) that recognize allogeneic HLA-C molecules on trophoblast. Where this results in excessive dNK inhibition, the risk of pre-eclampsia or growth restriction is increased....

Tissue Specific Heterogeneity in Effector Immune Cell Response

Post pathogen invasion, migration of effector T-cell subsets to specific tissue locations is of prime importance for generation of robust immune response. Effector T cells are imprinted with distinct "homing codes" (adhesion molecules and chemokine receptors) during activation which regulate their targeted trafficking to specific tissues. Internal cues in the lymph node microenvironment along w...

Effects of Dendritic Cell Vaccine Activated with Components of Lieshmania Major on Tumor Specific Response

Background: Dendritic cells (DCs) contribute essentially to the outset and course of immune responses. So in patients with malignancy, there have been considerable interests in use of these cells in different interventions. Objective: To evaluate the impact of Leishmania major’s components on DC maturation and their use as a therapeutic agent against t...

Proapoptotic Bim regulates antigen-specific NK cell contraction and the generation of the memory NK cell pool after cytomegalovirus infection

Apoptosis is critical for the elimination of activated lymphocytes after viral infection. Proapoptotic factor Bim (Bcl2l11) controls T lymphocyte contraction and the formation of memory T cells after infection. Natural killer (NK) cells also undergo antigen-driven expansion to become long-lived memory cells after mouse cytomegalovirus (MCMV) infection; therefore, we examined the role of Bim in ...

IL-2–dependent adaptive control of NK cell homeostasis

Activation and expansion of T and B lymphocytes and myeloid cells are controlled by Foxp3(+) regulatory T cells (T reg cells), and their deficiency results in a fatal lympho- and myeloproliferative syndrome. A role for T reg cells in the homeostasis of innate lymphocyte lineages remained unknown. Here, we report that T reg cells restrained the expansion of immature CD127(+) NK cells, which had ...
